MODIFIED XENOGRAFT MODELS TO ASSESS TUMOR INITIATION

PRE-TREATED CELL CULTURES ARE USED TO GENERATE TUMOR XENOGRAFTS IN IMMUNODEFICIENT MICE AND TUMOR FORMATION IS MEASURED IN TIME, COMPARED TO A VEHICLE PRE-TREATED CULTURE.  

Standard xenograft systems are first treated with a lead compound, then cells are extracted and inoculated to new animals, researchers can compare the increase in number of cells to determine tumor initiation frequency. This is the gold standard assay for cancer stem cell function.


In the second phase standard xenograft systems are then treated with current standard of care chemotherapy, residual resistant tumors are enriched in cancer stem cells, that are then subsequently xenografted into new animals where the activity of the lead being studied can be assessed.
